Change concentration sequence

New Turbidimetric assay (USP <81>/JP 4.02) documents have 5 concentrations of the standard, that is, 1 reference concentration, which is the median level, plus 4 extra concentrations. You can change concentration values, add or delete concentrations, and change the position of concentrations in the sequence.

Create a Turbidimetric assay (USP <81>/JP 4.02) document.

About this task

By default, the reference standard of Turbidimetric assay (USP <81>/JP 4.02) documents has a 5-step concentration sequence. You can view the default setup in the By Sequence editor. The data table has five rows for the five steps of the standard's concentration sequence and three columns for its three replicates:

In this example, you work with a 7-step sequence. Therefore, you need to add two concentration steps and adjust the values of the concentrations in the sequence.

Procedure

To change the concentration sequence:
  1. In the Content editor, go to Setup > Assay elements > Standard .
  2. Select Stepwise concentration sequence.
  3. Use the Creatable elements pane to add two more Concentration elements.
    Result: PLA 3.0 adds a sixth and a seventh concentration with the default value 1 to the sequence.
  4. Change the concentration values as shown in the following table.

    Tip: You can drag an element up and down the concentration sequence and drop it when the dark blue line highlights the position where you need the element.
    Result:
    The data table in the By Sequence editor adjusts automatically to reflect your changes. The row headers (gray column on the left) show the values you entered in the Content editor.

  5. Save the document.

Result

The Stepwise concentration sequence element in the Setup section of the Content editor should now have 7 steps. The reference concentration (median level of the standard) of this sequence is 100 µg/mL:
