By default, Cylinder plate assay (USP <81>) documents contain a single test sample. And the stepwise concentration sequence of the reference standard consists of 5 concentrations. The default settings therefore imply 15 plates (Petri dishes):

The standard has a 5-step concentration sequence by default and therefore requires 12 plates (Petri dishes), that is, 3 plate replicates for each of the 4 concentration steps.
The test sample requires 3 plates.
